Design: RNA was extracted from frozen endosperm samples (multiple endosperms dissected from seeds from the same fruit were pooled per sample) via RNAqueous Total RNA Isolation Kit (Invitrogen) according to the kit protocol. Libraries were prepared and sequenced at the MIT BioMicroCenter via NEBNext Ultra II Directional RNA Library Prep Kit for Illumina (polyA-based isolation). Samples were pooled and sequenced on one NovaSeq 6000 S4 flowcell (50bp, single end reads).

RNAseq and Enzymatic methyl sequencing of endosperm of reciprocal hybrids between Nymphaea thermarum and Nymphaea dimorpha, in order to characterize gene expression and DNA methylation patterning. The goal of this study is to identify genetic imprinting and parent-of-origin-specific DNA methylation patterning in endosperm of water lilies.
